While you may encounter some late season albacore on the way to the island, most of the fishing at Guadalupe is done with 40 and 50 pound line, preferably on a 2 speed. They will at times bite the 60 pound, but more than likely that will be when you are on the helium balloon or kite, with 80lb being preferred. The island is notorious for it's great white population so getting the fish to the boat as quickly as possible is key. The tuna can be touchy there so the use of flouracarbon at times can significantly increase your bites. I always bring balloons to fish bobber style when there are wind against current scenario's which allow you to fish areas otherwise inaccessable while flylining a bait.
There is also very good grade yellowtail there that can be targeted with 40 to 60 pound line by either flyling, using a sliding sinker to fish deeper in the water column or my favorite, the yo yo iron.
I would recommend having a 40lb 2 speed rig, a 50lb 2 speed rig as well as a 60lb 2 speed rig. With tuna caught to 100 pounds on a regular basis, I certainly would not recommend going lighter than 30lb while fishing the island, unless you just want to get taxed by Mr. Whitey.
The last two times I have been to guadalupe there has been absolutely NO reason to drop below 40lb. I fish 40lb on a 6465H with an Avet 2spd and its a good live bait combo. I also fish flouro leaders with circle hooks, and have been in many a debate about whether to fish flouro and whether to fish j hooks. I have heard tons of differing opinions, but I fish flouro with circle hook and do just fine.
I also fish 50-60lb on the yo-yo iron for the toad yellowtail there, and have had great success. Its really fun to watch everyone soaking dines (on the long soak) and getting no tuna bite, then you hang a big yellow on the iron. Watch how fast the switch, then its time to go back to chunking when everyone is yo-yo/pooper loopinig.
I also bring an 80lb 2speed rig with me. Use it on the kite, use it if we troll, use it pooper looping.
Surface iron - bring it, you might need it, but most of my experience at lupe is all tuna in the am/day, then pooper looping at night.
Now, on a 5 dayer, its possible you may swing in by San Martin on the way home, so bringing the lighter stuff may be useful there.

I know exactly what you mean Dave. Last year when everyone was soaking flylined dines' at outer rock waiting for a tuna bite, I decided to drop a blue and white Salas 6X to the bottom in 300 feet and got nailed on the way up on the first drop. Next thing you know I got 12 other guys dropping jigs all around me...

Well, you are going to the right place for slug yellows. The current world record came from Guadalupe. Bring the heavy iron for yo yo fishing (Salas 6X is my favorite) as well as torpedo sinkers for dropper looping at night. You may even get a few on a glow in the dark yo yo jig at night.

Tim is dead on about the Fluro. Bring plenty. The times that I have gone to Lupe, the fish have been finicky and those that weren't using it missed out. 40# test was the rule, but some guys did get bit on the 60#. Saw nothing heavier. The yellows are nasty there and expect to get rocked...ALOT. Bring lots of film, the island itself is amazing and the Discovery channel doesn't do the sharks justice. Be prepared, if the current isn't going it gets slow. But when it's on, it's on bigtime. The times I went, the night bite wasn't there, but I hear it can be great! Hope ya slay em!!
