alot of people put spectra on a reel as backing so they can fish heavier line on the reel without sacraficing capacity if they hook a big fish.
spectra is smaller diameter than mono for almost 250% the strength for the same size.
i put 300 yards of 50 pound spectra and 100 yards of mono over the spectra on my sx connected with a bimini/ albright knot. combined together the reel has 400 yards of line on it

Depends what you want to do with the reel. Ovbiously the 2 speed has more versatility. I like my 2 speed for big swim baits because i can vary the line retrieve speed while cranking the handle the same. The advantage to the single speed is the stock handle can be replaced with a counter balenced handle that makes the reel all the more balenced.
I would buy a single speed sx: for local stuff fill it with strait 20 mono (thats plenty mono). for long range, think about spectra
